چکیده

Enzymatic attachment of biotin to proteins requires the interaction of a distinct domain of the acceptor protein (the “biotin domain”) with the enzyme, biotin protein ligase, that catalyzes this essential and rare post-translational modification. Both biotin domains and biotin protein ligases are very strongly conserved throughout biology. This review concerns the protein structures and mechanisms involved in the covalent attachment of biotin to proteins. J. Nutr. 129: 477S–484S, 1999.

Production of Recombinant Streptavidin and Optimization of Refolding Conditions for Recovery of Biological Activity

Background: Streptavidin is a protein produced by Streptomyces avidinii with strong biotin-binding ability. The non-covalent, yet strong bond between these two molecules has made it a preferable option in biological detection systems. Due to its extensive use, considerable attention is focused on streptavidin production by recombinant methods. Methods: In this study, streptavidin was express...

The 1.3 A crystal structure of a biotin-binding pseudoknot and the basis for RNA molecular recognition.

A pseudoknot-containing aptamer isolated from a pool of random sequence molecules has been shown previously to represent an optimal RNA solution to the problem of binding biotin. The affinity of this RNA molecule is nonetheless orders of magnitude weaker than that of its highly evolved protein analogs, avidin and streptavidin. To understand the structural basis for biotin binding and to compare...

Anti-biotin antibodies offer superior organelle-specific labelling of mitochondria over avidin or streptavidin.

A number of endogenously biotinylated proteins are found in both cytosol and mitochondria of mammalian cells from many tissues, including liver, spleen, pancreas, kidney, and intestine. Therefore, caution should be taken when using the biotin detection system. Endogenous biotin can interfere with staining systems that employ the use of biotin-avidin- or biotin-streptavidin-based detection syste...

Symposium: Nutrition, Biochemistry and Molecular Biology of Biotin Cellular Uptake of Biotin: Mechanisms and Regulation

This review describes our knowledge of biotin transport in the small intestine of humans and other mammals and presents recent findings in the area. Previous studies have shown that biotin transport across the brush border membrane of the small intestinal absorptive cells occurs via a carrier-mediated, Na gradientdependent, electroneutral mechanism. Exit of biotin out of the enterocyte, i.e., t...

Plant sucrose-H+ symporters mediate the transport of vitamin H.

A cDNA coding for a vitamin H (biotin) transport protein from Arabidopsis was identified by genetic complementation of a biotin uptake-deficient yeast mutant. Vitamin H transport by this protein was sensitive to the SH-group inhibitor p-chloromercuribenzene sulfonic acid (PCMBS) and to the uncoupler carbonyl cyanide-m-chlorophenylhydrazone (CCCP), suggesting an energy-dependent biotin-H+ sympor...
